Gliomas are the most common primary brain tumors. Presence of promoter methylation, which causes epigenetic silencing of the MGMT gene, a DNA repair gene on chromosome 10q26, is a positive predictive factor for chemotherapeutic response to alkylating agents in the treatment of gliomas. The methylation status of the MGMT promoter is determined by methylation-specific PCR analysis (MSP). The DNA is amplified with primers specific for the methylated and the unmethylated product. |
