This assay detects loss of glycosyl phosphatidylinositol (GPI)-anchored proteins on erythrocytes and neutrophils, and is of diagnostic utility in suspected cases of paroxysmal nocturnal hemoglobinuria (PNH). Glycophorin and CD15 are used to assist in gating erythrocytes and neutrophils, respectively. Expression of CD55, CD59, CD16 and CD66b is analyzed, with the goal of detecting and quantifying abnormal populations with absent or dim GPI-linked antigen expression. |
